چکیده

This study reports on the development of a novel impedimetric immunosensor design using plant-derived antigenic glycoprotein for detection dengue virus (DENV) IgG antibodies. The electrochemical platform was constructed screen-printed carbon electrode (SPCE) modified with graphene/titanium dioxide (G/TiO2) nanocomposite to improve in terms performance and specific surface area. A envelope domain III (EDIII) protein used as probe this immunosensing strategy. Under optimised sensing conditions, demonstrated high sensitivity towards DENV wide linear working range (62.5–2000 ng/mL), limit 2.81 ng/mL. showed specificity discriminating against antibodies other infectious disease, including closely related Zika (ZIKV). reliability serological diagnosis verified by challenging serum samples, compared conventional enzyme-linked immunosorbent assay (ELISA). As shown its remarkable throughout study, devised is proposed reliable practical diagnostic tool realistic applications.

Neutralizing Antibody Response and Efficacy of Novel Recombinant Tetravalent Dengue DNA Vaccine Comprising Envelope Domain III in Mice

Background: Dengue is a global arboviral threat to humans; causing 390 million infections per year. The availability of safe and effective tetravalent dengue vaccine is a global requirement to prevent epidemics, morbidity, and mortality associated with it. Methods: Five experimental groups (6 mice per group) each of 5-week-old BALB/c mice were immunized with vaccine and placebo (empty plasmid) ...

Evaluation of envelope domain III-based single chimeric tetravalent antigen and monovalent antigen mixtures for the detection of anti-dengue antibodies in human sera

BACKGROUND Flavivirus cross-reactive antibodies in human sera interfere with the definitive identification of dengue virus (DENV) infections especially in areas with multiple co-circulating flaviviruses. Use of DENV envelope domain-III (EDIII) can partially resolve the problem. This study has examined the effect of (i) incorporating the EDIIIs of four DENV serotypes into a single chimeric antig...

Characterization of a dengue type-specific epitope on dengue 3 virus envelope protein domain III

Dengue virus (DENV) is a mosquito-borne disease caused by four genetically and serologically related viruses termed DENV-1, -2, -3 and -4. The DENV envelope (E) protein ectodomain can be divided into three structural domains designated ED1, ED2 and ED3. The ED3 domain contains DENV type-specific and DENV complex-reactive antigenic sites. To date, nearly all antigenic studies on the E protein ha...
